ACE-031 is a recombinant fusion protein consisting of the extracellular domain of human activin receptor type IIB (ActRIIB) fused to the Fc portion of human IgG1. It functions as a soluble decoy receptor, binding and neutralizing myostatin and related TGF-β superfamily ligands that normally inhibit muscle growth. By sequestering these negative regulators, ACE-031 promotes muscle hypertrophy and increased strength. Originally developed by Acceleron Pharma for Duchenne muscular dystrophy, clinical trials showed significant increases in lean body mass and thigh muscle volume. However, development was paused due to safety signals including nosebleeds and gum bleeding.
