Triptorelin is a synthetic decapeptide analog of gonadotropin-releasing hormone (GnRH/LHRH) with significantly greater potency and longer duration of action than natural GnRH. As a GnRH agonist, triptorelin initially causes a surge in LH, FSH, and sex hormones ("flare effect"), followed by downregulation of GnRH receptors and profound suppression of the reproductive axis with continuous administration. Triptorelin is FDA-approved for advanced prostate cancer, endometriosis, uterine fibroids, precocious puberty, and as part of fertility treatment protocols. Depot formulations allow once-monthly or every-3-month dosing.
